摘  要:目的 探讨重组人转化生长因子-β1 (rhTGF- β1 )对成人骨髓间充质干细胞(MSCs)分化为成骨细胞的影响。方法 体外分离、扩增成人骨髓MSCs。采用不同浓度的rhTGF= β1 诱导成人骨髓MSCs体外分化为成骨细胞。利用倒置光学显微镜、透射电镜、四甲基偶氮唑盐比色、碱性磷酸酶(ALP)染色、ALP活性测定等方法研究成人骨髓MSCs增殖和分化情况。结果 成人骨髓MSCs的增殖及分化与rhTGF =β1呈剂量依赖关系,低浓度时促进增殖,高浓度抑制增殖, 5μg/L浓度时增殖达到高峰。其浓度升高促进MSCs分化。结论 成人骨髓MSCs分化为成骨细胞,rhTGF= β1 浓度为5μg/L时较合适。Objective To investigate the effect of recombinant human transforming growth factor-β_1(rhTGF-β_1) on the differentiation from the adult human bone marrow mesenchymal stem cells (MSCs) to osteoblast.Methods Separation and proliferation of the MSCs were conducted.MSCs from the adult human bone marrow was induced into osteoblast with different levels of rhTGF-β_1.Proliferation and differentiation of adult human bone marrow MSCs were examined by phase-contrast microscope,transmission electron microscope,MTT,alkaline phosphatase(ALP) staining and the determination of ALP activities.Results The proliferation and differentiation of adult bone marrow MSCs were dose-dependently related with the concentration of rhTGF-β_1.Low concentration of rhTGF-β_1 promoted the proliferation of the adult human bone marrow MSCs and vice verse,which peaked at 5ug/L.The increased concentration of rhTGF-β_1 could promote MSCs differentiation.Conclusion 5 μg/L of rhTGF-β_1 is suitable for the adult human bone marrow MSCs to differentiate into osteoblast.
