摘  要:目的探讨重组水蛭素(rH)对大鼠冠状动脉血栓形成的抑制作用。方法将160只SD大鼠随机分为8组,每组20只,实验组1~4分别静脉注射rH1.000,0.500,0.250,0.125mg·kg1;实验组5静脉注射Refludan,0.500mg·kg1;实验组6静脉注射肝素2000U·kg1;模型组静脉注射等量0.9%氯化钠注射液;对照组不给药。10min后将大鼠麻醉开胸,将吸有50%三氯化铁溶液的滤纸片贴于实验组1~6和模型组大鼠的冠状动脉前降支根部,60min后取下,以制备大鼠冠状动脉血栓模型。对照组大鼠冠状动脉前降支根部敷贴不含三氯化铁溶液的滤纸片。考察各组大鼠心肌缺血面积评分、左心室伊文思蓝含量、心肌超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量,并做心肌病理形态学检查。结果实验组1~5大鼠心肌缺血面积评分分别为12,16,24,28,19分,均明显低于模型组(39分)(均P<0.01);实验组1~3和实验组5,6心肌伊文思蓝含量明显低于模型组(均P<0.01);实验组1心肌SOD活力[(54.64±12.05)NU·mg1]明显高于模型组[(30.46±8.73)NU·mg1](P<0.01),实验组2,3,6心肌SOD活力亦高于模型组(均P<0.05);实验组1,2,5心肌MDA含量明显低于模型组(均P<0.01),实验组3,6亦低于模型组。病理学检查可见实验组3,4大鼠冠状动脉有血栓形成,实验组2血栓形成明显减轻。结论受试rH具有明显?Objective To explore the antithrombotic effect of recombinant hirudin in rats. Method We randomized 160 SD rats into eight groups, twenty rats in each group. Group 1, 2, 3 and 4 received rH 1.000, 0.500, 0.250, 0.125 mg·kg -1 ; group 5 received refludan 0.500 mg·kg -1 ; group 6 received heparin 2 000 U·kg -1 ; the model group receive the same volume of sodium chloride injection, i.v., and the control group received nothing. After 10 min, filter paper marinated in 50% FeCl 3 solution was attached to the low front part of the coronary artery for 60 min in group 1, 2, 3, 4, 5, 6 and in the model group to induce thrombosis. The control group were operated in the same way but had no FeCl 3. The antithrombosis effect of rH was evaluated by scores of infarcted area, Evans Blue content of left ventricle myocardium, difference of SOD activity and MDA content. Results The scores of infarcted area in group 1, 2, 3, 4 and 5 were respectively 12,16,24,28,19, which were all significantly lowered than 39 in the model group. Evans Blue content in group 1, 2, 3, 5 and 6 was significantly lowered than in control group (P<0.01). The SOD activity of myocardium in group 1 was(54.64±12.05) NU·mg -1 , significantly higher than(30.46±8.73) NU·mg -1 in the model group (P<0.01).The SOD activity of myocardium in group 2, 3 and 6 was also significantly higher than in the model group (P<0.05). The MDA content in group 1, 2, 3, 5 and 6 was significantly lowered than in the model group (P<0.01). Coronary artery thrombosis was showed in group 3, 4 by pathological examination. Coronary artery thrombosis in group 2 was obviously reduced. Conclusion rH has an antithrombotic effect which is dose-dependent.
