摘 要:目的考察纳米羟喜树碱注射用灭菌粉末经静脉注射给药后在家兔体内的药动学特征。方法建立测定家兔血浆中羟喜树碱含量的高效液相色谱法,标准曲线的回归分析采用加权最小二乘法;采用3p97药动学计算软件模拟房室模型并计算纳米羟喜树碱注射用灭菌粉末在家兔体内的药动学参数。结果纳米羟喜树碱注射用灭菌粉末在家兔体内的代谢符合1/C2权重的三室模型。相同剂量下普通羟喜树碱在家兔体内的半衰期较纳米制剂短。结论纳米羟喜树碱注射用灭菌粉末的药动学特性与普通羟喜树碱制剂的特性有明显区别。Objective To study the concentration and pharmacokinetic parameter after the injection of hydroxycamptotheinum (HCPT) nanoparticles in rabbits. Method HPLC method was established for the determination of HCPT in rabbit plasma; The pharmacokinetic calculation software 3p97 was used to simulate the compartment model and to calculate the pharmacokinetic parameters. Results The nanoparticle injection powder was consistent to three-compartment model of 1/C 2 weight; the half life (t 1/2β)was 43.114 min at the dosage of 5.0 mg·kg-1 ; C max was 3.27 mg·mL -1 ; AUC was 50.65. Conclusion The nanoparticle injection is superior significantly than the general injection powder in pharmacokinetic characteristics.
