摘 要:利用异源的单克隆抗体和多克隆抗体通过间接酶联免疫吸附试验,检测了深圳唐菖蒲的叶组织、块茎和试管苗中的病毒。单克隆抗体检测效果优于多克隆抗体,并与电镜观察结果相符。在间接酶联免疫吸附试验中,同源多克隆抗体对烟草花叶病毒(TMV)检测的灵敏度比侵染性试验高30倍。同时对该方法在大规模检测其它无病毒试管苗中的重要性和可行性进行了讨论。The heterologous monoclonal and polyclonal antibodies were used to detect the existence of viruses, the rod shaped (TMV-related) and thread shaped (PVY-related) in leaves, corm and plantlets of Gladiolus (Gladiolus hybridious Hort) from Shen Zhen by indirect ELISA. For detection of the rod shaped, the TMV common strain specific monoclonal antibody used in the indirect ELISA gave much better sensitivity than its polyclonal antibody. The results of detection are coincident with electron microscopy. The sensitivity was also checked between the indirect ELISA with homologous polyclonal antibody to TMV and the differential host (Nicotiana tobacum var. Xanthi-nc) infectivity, the former was about more 30 fold than the latter one. The important and feasibility of the method presented in this report for detection other kinds of virus-free plantlets in large scale had been discussed.
