摘  要:目的观察1,25-(O H)2D对阿霉素肾病大鼠血管紧张素转化酶(AC E)表达的影响。方法1283只周龄相同体重相近的雄性Sprague D aw ley(SD)大鼠随机分为4组;正常对照组、肾病组、激素治疗组和激素+阿法D治疗组(简称联合组)。尾静脉注射阿霉素制备大鼠阿霉素肾病模型,实验当天、第14天、第283天及第42天每组随机抽取8只大鼠取血、尿与肾脏,测血中BU N、C r、A lb、C H O、C a、P、A LP的浓度,24h尿蛋白排泄量血浆与肾皮质AC E浓度,病理积分评价大鼠肾脏损害程度。结果实验初,各组大鼠血中BU N、C r、A lb、C H O、C a、P、A LP的浓度、24h尿蛋白排泄无明显差异,自实验第14天起,肾病组、激素组与联合组大鼠血BU N、C r、C H O、A LP含量、24h尿蛋白排泄量较正常组增高,A lb、C a、P含量较正常组降低(P<0.01)。自实验14天起,激素组与联合组C a、P浓度高于肾病组(P<0.01),而联合组高于激素组(P<0.01)。血中A C E浓度无明显变化,肾局部AC E浓度经激素治疗后明显受抑,但联合治疗效果更显著(P<0.01)。实验末,肾脏病理积分依次为肾病组、激素组、联合组、正常组。相关分析发现:血A lb、C a、P浓度与病理积分呈负相关(P均<0.01),肾皮质AC E、BU N、C r浓度与病理积分呈正相关(P<0.01),血C a、P浓度与肾皮质A C E浓度呈?To observe influence of 1, 25-(OH)2D3 on angiotension invertase of rat with nephropathy. 128 male Sprague Dawley (SD) rats with the same weeks old and near weight were randomly divided into 4 groups: normal control group, nephropathy group, treated group with hormone, treated group with hormone+α-D3 (for short combine group). Preparation for nephropathy mode by injection of adrianycine via caudal vein, on the day, 14th, 28th and 42nd day of trial randomly selected 8 rats and respectively collected blood, urine and kidney to detect the concentration of BUN, Cr, Alb, CHO, Ca, P, ALP in blood, the output of urinary protein in 24 hr and the concentration of ACE in plasma and renal cortex and the injury degree of kidney. On the initial stage of trial, concentration of BUN, Cr, Alb, CHO, Ca, P, ALP, ACE and output of urinary protein in 24 hr in the blood or urine of four groups of rats without obvious difference. On the 14th day, content of BUN, Cr, CHO, ALP and output of urinary protein in 24 hr in the blood of each group of rats was higher than that in normal group of rats, but content of Alb, Ca, P was lower than in normal group, with significant difference (P <0.01); from the 14th day of trial, Concentration of Ca, P of groups with hormone and combined drug was higher than that in nephropathy group (P <0.01), that in combined group was higher than that in hormone group (P <0.01). Concentration of AEC in blood, no obvious changes, local concentration of ACE in kidney was markedly inhibited, after treated with hormone, but effect with combined drug was more significant (P <0.01). On the trial end, pathological score of kidney was successively nephropathy, hormone, combined and normal group. Related analysis showed that the concentration of Alb, Ca, P with pathological score was negative relation ( All P <0.01), the concentration of ACE, BUN, Cr with pathological score was positive relation (all P <0.01), the concentration of Ca, P in blood with that of ACE in renal cortex was negative relation (all P <0.01).
