摘  要:目的研究用鸡卵黏蛋白为配基亲和色谱法分离纯化麦胚凝集素,并对纯化的麦胚凝集素进行性质分析。方法先制备鸡卵黏蛋白的亲和吸附剂,然后用其纯化麦胚凝集素。结果此亲和色谱方法使纯化后的麦胚凝集素比其粗提液纯度提高约10 0 0倍,相对分子质量为2 0 4 2 0 ,pI为5 .10。PurposeTo make some research on the method of purification of wheat germ agglutinin (WGA) using Sepharose-4B which is combined with gallinaceous ovomucoid as an affinity adsorbent, and to analyze its properties.MethodsChromatographic column coupled with gallinaceous ovomucoid was used to isolate and purify WGA.ResultsThe hemagg1utinating activity of purified WGA was raised by about 1 000 times. The molecular weight of WGA was 20 420, and its isoelectric point is 5.10.ConclusionThis means of affinity chromatography with gallinaceous ovomucoid has been proven to be a kind of economical and applicable method to purify and isolate WGA.
