摘　　要：目的探讨人肺腺癌细胞(SPC-A-1)70 KD热休克蛋白的提取方法及体外抗肿瘤作用.方法 SPC-A-1培养至对数生长期,43 ℃加热处理12 h,超声破碎细胞离心取上清、盐析沉淀、30 KD分子筛滤过、SDS-PAGE分离鉴定,MTT法测定其体外细胞毒活性.结果提取产物经SDS-PAGE分析,其电泳区带在70 KD位置处,细胞毒性试验表明,此复合物能够加强脐血有核细胞的抗肿瘤效果.结论该提取方法流程简便、实用,有助于临床广泛开展热休克蛋白的应用.ObjectiveTo study the isolated and purified method of 70KD heat shock proteins-antigen complex(HAC) from spc-A-1 cells.MethodsSpc-A-1 cells were cultured until logarithmic growth, and then heat shock treatment with (43 ℃) for 12 hours, (70 KD)HAC was purified from the medium by ultrasonic shatter, centrifugation filtration, salting precipitation with ammonium sulfate, isolated with SDS-PAGE.ResultsThe isolated proteins were confirmed to be a sample with MW 70 KD by SDS-PAGE, and proved to be higher biological activity by CTL test in vitro.ConclusionThe procedure is simple and does not need complicated treatment.
