摘  要:目的 探讨超声心动图评价骨髓间质干细胞移植对心肌梗死后左心室收缩功能的作用及治疗价值。方法 24只新西兰白兔随机分为干细胞移植组和对照组。结扎左冠状动脉回旋支建立心肌梗死模型后,移植组与对照组心肌梗死区中央及周边分别注射骨髓间质干细胞和DMEM培养液。应用超声心动图对结扎前、心肌梗死后3d及注射移植后4周左室大小和收缩功能指标进行检测。结果 实验期内两组各有1只兔死亡。干细胞移植后4周,移植组左室射血分数,后壁的收缩幅度、增厚率和峰值收缩速度以及二尖瓣环6个部位的平均峰值收缩速度均较梗死后3d时显著增加;移植组左室腔径明显小于对照组,收缩功能指标明显高于后者。结论 自体骨髓间质干细胞移植能有效改善兔心肌梗死后左室收缩功能,减轻室腔扩大。Objective To investigate the effects of autologous bone marrow stromal stem cells(MSSCs) transplantation on cardiac function in acute myocardial infarction(AMI). Methods Twenty four New Zealand white rabbits were divided into two groups randomly, control group and MSSCs group. The circumflex branch of left coronary artery was ligated to produce the myocardial infarction model. After infarction, MSSCs group was injected with autologous MSSCs, while control group was injected with DMEM culture medium. The size and systolic function of left ventricle (LV) were measured by echocardiography before and 3 days after infarction, 4 weeks after implantation. Results One rabbit died unexpectedly in each group during the study. Four weeks after implantation, LVEF, systolic displacement, thickening rate and peak systolic velocities of LV posterior wall, and mean peak contractive velocities in six parts of mitral annular increased significantly than 3 days after infarction in the MSSCs group. Compared with the control group, the LV diastolic diameter was smaller and the contractile function was better in the MSSCs group. Conclusions Autologous bone marrow stem stromal cells transplantation can improve systolic function and alleviate the enlargement of infarcted LV.
