摘 要:目的研究脐血CD34+HPC体外扩增诱导产生功能性树突状细胞(DCs)的有效方案。方法Ficoll分离脐血获得单个核细胞,免疫磁珠阳性分选CD34+HPC,3H—TdR检测DCs激发异体T细胞增殖的能力,MTT法检测特异性CTL的杀伤能力。结果SCF+FL+GM-CSF+IL-4诱导生成的DCs激发T细胞增殖和特异性CTL的杀伤能力均高于其他组。结论SCF+FL+GM-CSF+IL-4可有效扩增CD34+HPC并诱导产生功能性DCs。Objective To probe the effective program of expansion and function of dendritic cells (DCs) derived from human cord blood CD34+ hematopoietic precursor cells(HPC). Methods The isolation of CD34 + cells from human cord blood was used with mini-MACS. The proliferation of T cells activated by allo-DCs was measured with 3H-TdR incorporation. The cytotoxic activity of special CTL was measured with MTT. Results The proliferation of T cells and the cytotoxic activity of CTL stimulated by DCs inculture with SFG4 were higher than those in other groups. Conclusion CD34 + HPC could expand effectively and be induced into DCs with the combination of SCF, Flt-3 Ligand, GM-CSF and IL-4.
