摘 要:目的探讨不同细胞因子组合对脐带血AC133+细胞体外扩增效率的影响。方法采用免疫荧光标记法,使用流式细胞仪测定新鲜分离的、不同培养条件下体外培养7、14d的脐带血AC133+细胞的扩增效率。结果新鲜脐带血AC133+细胞的比例为(0.93±0.2)%。短期液体扩增培养后,脐带血AC133+细胞的比例发生明显变化,其中在细胞因子SCF、FL和IL-3组合下,AC133+细胞的细胞数及扩增倍数与SCF、FL和IL-11组合相比较,差异非常显著(P<0.01)。结论在体外短期扩增培养中,早期效应细胞因子能显著上调脐带血AC133+细胞的比例;用SCF、FL和IL-3组合扩增脐带血造血干/祖细胞,能够获得更高的体外扩增效率。Objective To investigate the effects of different cytokine combinations on the ex-vivo expansion of umbilical cord blood AC133 + cells. Methods The expansion efficiency of the umbilical cord blood AC133 + cells which were freshly isolated and incubated in serum-free liquid culture system for 7 d and 17 d with different cytokine combinations in vitro were analyzed by flow cytometric im-munoflurescence methods. Results Frequency of AC133+ cells in the freshly isolated umbilical cord blood mononuclear cells only was a significantly low (0.93% ±0.2 SEM). After short-term incubation in culture system with cytokines in vitro, the percentage of AC133+ cells increased. The total number and expansion was remarkably higher in the presence of IL-3, FL and SCF combination than that in the presence of IL-11, FL and SCF combination. There were significant difference between the two groups (P<0.01). Conclusion During short-term culture in vitro, the early-effective cytokines can up-regulate the percentage of AC133+ cells. In the presence of IL-3, FL and SCF combination, we may gain more expansion efficiency of the umbilical cord blood AC133+ cells than that in IL-11, FL and SCF combination.
