摘  要:目的 探讨β3肾上腺素能受体(β3AR)基因多态性及环境因素(看电视)与儿童体重增加的关系。方法 选取大庆地区400名儿童进行5年随访,入选时和第5年分别测定身高、体重,检测β3AR基因多态性,以多因素分析探讨各基因型与儿童体重增长的关系及此基因与环境因素(看电视)对儿童体重增长的共同作用。结果 (1)调整年龄、性别因素的影响后, 5年后β3AR基因第64位TA/AA基因型组体重高于TT基因型组[ (38. 3±10. 5)kgvs(36. 3±8. 3)kg,P<0. 05];逐步回归分析显示, 5年后体重与随访前体重、看电视时间、β3AR基因TA/AA型正相关。(2)将每周看电视时间分为3组(均值分别为2. 3、4. 3、7. 3h), 5年的体重增长为(12. 6±0. 6)kg、(13. 8±0. 4)kg、(15. 9±0. 5)kg(第1、3组间P<0. 01);在3组内不同基因型间进行比较,每周看电视时间为7. 3h组,TA/AA基因型体重增值明显高于TT基因型(P<0. 05)。结论 (1)β3ARTA/AA基因型与儿童体重增长密切相关。(2)环境因素(看电视时间)可加强β3ARTA/AA基因型对体重增加的作用。Objective To investigate the association of β_ 3-adrenergic receptor (AR) gene polymorphism and environment factors (TV-watching) with weight gain in children. Methods Four-hundred children in elementary schools in Daqing were enrolled in the present study. Body height and weight were measured in 1 st, 5th year. The polymorphism of β_ 3-AR gene was detected by PCR-RFLP. The relationship between β_ 3-AR gene polymorphism and weight gain and the effect of genetic and environmental factors (TV-watching) on weight gain were analysed by multivariate regression analysis with SAS software package. Results (1) After adjustment for age and sex, the body weight of 5th year of TA/AA genotype group was significantly higher than that in TT group \[(38.3±10.5)kg vs (36.3±8.3)kg, P<0.05\]. Stepwise regression analysis showed that the body weight of 5th year was positively correlated with baseline body weight, the time of TV-watching, TA/AA genotype. (2) When stratified all the subjects to threegroups(tertiles)bytimeof TV-watching (mean time was 2.3, 4.3 and 7.3 h respectively), the weight gain were (12.6±0.6)kg、(13.8±0.4)kg、(15.9±0.5)kg, the P value between 1st and 3 rd group was <0.01. Compared the weight gain between genotypes in three groups, weight gain of TA/AA genotype was significantly more than that of TT genotype (P<0.05) group in which the time of TV-watching was 7.3 hours per week. Conclusions (1) β_ 3-AR gene polymorphism is significantly correlated with weight gain. (2) Environmental factors (time of TV-watching) enhance the effect of the genotype of β_ 3-AR TA/AA in promoting weight gain.
