摘 要:【目的】研究丹参酚酸B对人(K562)细胞系生长的抑制效应,并探讨其作用机制。【方法】用不同浓度的丹参酚酸B加入体外培养K562细胞中,观察加药后细胞生长数量及其形态的变化;用MTT法检测丹参酚酸B的细胞毒作用;用Hoechest33342和PI双荧光染色法检测细胞凋亡。【结果】丹参酚酸B明显抑制K562细胞生长;IC50值为2.5×10-5mol/L;其抑制率与药物浓度呈剂量依赖关系;凋亡细胞的比例与药物浓度及作用时间呈正相关。【结论】丹参酚酸B能有效抑制K562细胞的增殖。To study the effects and mechanism of Salivanol acid B on K562 cell line.Salivanol acid B of different concentrations were added into the medium in which the cells were cultured. The changes of cell morphology were observed by phase contrast microscope. Cellulotoxic activity of Salivanol acid B was defected by MTT. The number of apoptotic cells was quantitatively assessed by fluorescent light microscope.The growth of K562 cell line was remarkably inhibited by Salivanol acid B. The IC_(50) values of Salivanol acid B for K562 cells was 2.5×10^(-5) mol/L. The inhibited rate and the rate of apoptotic cells were related with the medicine concentration.[Conclusion]Salivanol acid B can effectively inhibit the multiplication of K562 cell line.
