摘 要:目的:比较常规试剂盒和使用基因释放剂提取结核杆菌热休克蛋白70 (hsp70 )DNA模板的质量,为进一步获得目的基因优化方案。方法:提取结核杆菌DNA阶段,分别使用常规试剂盒和基因释放剂,然后采用相同的PCR方案进行扩增,溴化乙锭染色,紫外光下观察目的基因扩增效果。结果:常规试剂盒提取结核杆菌DNA ,未能扩增出相应的目的基因;而使用基因释放剂,则扩增效果良好。结论:常规试剂盒相比,基因释放剂在提取结核杆菌DNA时,能够制备良好的模板,为PCR扩增目的基因奠定基础。Objective: To Compare the qu a liy of the DNA templates of hsp 70 extracted from M. Tuberculosis with tradition al kit or genereleaser. Methods: The extracted DNA from M. Tuberculosis with traditional kit or genereleaser were subjected to the same PCR program, and then the PCR products were observed under ultra violet rays. Results: The target gene fragment was not seen when the te mplate was prepared with traditional kit, while it was clearly observed when pre pared with generaleaser. Conclusions: Genereleaser is he lpful in preparing higher quality temaplate from M. Tuberculosis compared with t radional kit.
