摘 要:目的证明白蚊伊蚊亲代能够经卵传递登革2型病毒给子代,并且在传递过程中病毒毒力呈逐渐增强趋势。方法采用套式PCR分批检测感染雌蚊的子一代至子四代卵内登革病毒结合C6/36细胞培养分离病毒及TCID50法滴定病毒滴度。结果白蚊伊蚊能经卵传递DEN-2,至少可传四代以上;且在传代中病毒毒力有增强的趋势。结论实验证明白蚊伊蚊对登革2型病毒有较大的媒介效能,且在维持登革病毒的自然循环中起重要作用。Objective To demonstrate that DEN 2 virus can be transmitted vertically to offsprings from parents infected with Aedes albopictus and its virulence tend to rise during the transovarial transmission. Methods Nest ploymerase chain reaction (Nest PCR) was employed to detect viruses in batches from the first generation eggs to the fourth, combined with virus isolation in cultured C6/36 cells and virus titer monitoring by 50 %tissue culture infection dose(TCID50). Results It showed that dengue virus could be transmitted at least to the fourth generation after the female infected with Aedes albopictus and there was a tendency of increased virulence from parent generation to offspring generation. Conclusion The experiment demonstrated that Aedes albopictus were capable of transmitting DEN 2 virus and had a potential role in the natural circulation of the virus.
