摘 要:目的了解海南省恶性疟原虫Pfmdr1基因中的关键性点突变是否与海南株恶性疟原虫产生氯喹抗性有关。方法采用套式PCR法分别扩增Pfmdr1基因中含有第86位和1246位氨基酸的多态性片段,并对扩增产物进行限制性内切酶酶切分析,观察是否存在突变位点。结果45个样本中,Pfmdr1基因第86位氨基酸均未发生点突变,即86位密码子是Asn而非突变型的Tyr;Pfmdr1基因发生D1246Y突变的样本有3个,其中1个是抗性株,2个为敏感株。结论我国海南株恶性疟原虫产生氯喹抗性与Pfmdr1基因的多态性无显著关系。Objective To identify whether point mutations existed in Pfmdr1 genes of Plasmodium falciparum from Hainan province and its relations with chloroquine resistance. Methods Nested PCR was applied to amplify fragments of Pfmdr1 genes including N86Y and D1246Y mutations. PCR products were digested with different restriction endomucleases to detect whether point mutations existed in Pfmdr1 genes. Results The Pfmdr1 gene N86Y mutation was not detected in all the 45 isolates but D1246Y mutation was found to be present in 3 of 45 isolates. One was chloroquine resistant and the other two were chloroquine sensitive. Conclusion Polymorphisms of Pfmdr1 gene seemed to have no apparent association with chloroquine resistance.
