摘  要:目的研究SO2衍生物对小鼠肝细胞DNA的影响。方法运用单细胞凝胶电泳技术(又称彗星试验)对小鼠进行腹腔注射,三个染毒组SO2衍生物剂量为125,250,500mg/kg体重,对照组注射生理盐水,并测量SO2衍生物引起小鼠肝细胞DNA损伤的尾矩(OTM)。结果雄性小鼠肝细胞DNA的OTM值分别为0.95,4.99,9.83,15.50,雌性小鼠肝细胞DNA的OTM值分别为0.81,3.85,6.82,12.87,这提示SO2衍生物可引起小鼠肝细胞DNA损伤,且随SO2衍生物注射剂量的增加DNA损伤加重,并有明确的剂量效应关系(r=0.993~0.996,P<0.001)。另外SO2体内衍生物对雄性小鼠肝细胞DNA损伤比雌性小鼠严重。结论这些结果意味着SO2体内衍生物具有引起哺乳动物肝细胞DNA突变的潜在危险。Objective To study the damage effects of sulfur dioxide (SO_2) derivative injection on DNA of liver cells from mice. Methods With the single cell microgel electrophoresis technique (comet test), three groups of 6 mice each received an intraperitionneal (i.p.) dose of SO_2 derivatives (125,250,500 mg/kg body weight), one negative control group of 6 mice each received i.p.0.2ml of saline, and Olive tail moment (OTM) in the cells was measured. Results The results showed OTM of nuclear DNA of liver cells from male mice were 0.95,4.99,9.83,15.50, and female mice were 0.81,3.85,6.82,12.87, respectively. It was suggested that SO_2 derivatives caused significant increases of OTM in the cells from liver tested in a dose-dependent manner(r=0.993~0.996,P<0.001).The results also indicated DNA damage of the liver cells from male mice was more serious than that from female mice, which remains to be further studied. Conclusion SO_2 pollution is a potential risk to damage genetic material DNA of liver cells from mammalian animals.
