大鼠肾小球系膜细胞产生的生长抑制性因子－转化生长因子β

摘　　要：本文观察了肾小球系膜细胞产生的自分泌生长抑制因子的情况。结果显示，肾系膜细胞培养上清中含有抑制自身生长的活性物质，后者有耐酸性；ＨＰＬＣ分析洗脱液中最主要的抑制性活性物质的分子量为１６～３０ｋｄ；抗－转化生长因子β（ＴＧＦβ）抗体能中和其大部分抑制活性；生物学方法检测表明上清中含有活性ＴＧＦβ；此外，培养液中加入抗ＴＧＦβ抗体能明显增强细胞的增殖。结果表明：大鼠肾系膜细胞能产生活性ＴＧＦβ，后者是一种自分泌的生长抑制性因子。The growth inhibitory factors produced by cultured glomerular mesangial cells of rat were investigated.The results showed that supernatants obtained from cultured mesangial cells contained growth inhibitory activity and the activity was resistance to acid treatment.HPLC analysis demonstrated peak inhibitory activity was resided in fractions with an estimated molecular weight range of 16-30kd.The activity was partially blocked by anti-TGFβ antibody.The presence of TGFβ was confirmed using the bioassay system.Furthermore,the addition of anti-TGFβ antibody directly into the culture media signifi.cantly enchanced mesangial cell proliferation.These results demonstrated that the mesangial cell produce active TGFβ,which functions as an autocrine growth inhibitor for mesangial cells.
