摘 要:目的通过观察排尿频率、膀胱内压和脑桥排尿中枢神经递质多巴胺(DA)含量的变化探讨针刺抑制左旋多巴(L-多巴)诱发的大鼠膀胱机能亢进的神经生物学机制.方法以在卡比多巴抑制外周多巴脱羧酶的前提下,腹腔注射的左旋多巴在中枢脱羧成多巴胺兴奋脑桥排尿中枢引起膀胱机能亢进的动物为模型,在动物清醒和自由活动状态下,应用连续膀胱测压技术动态观察电针对排尿频率和膀胱内压的影响.应用微透析和高效液相色谱-电化学检测技术,观察、分析电针后脑桥排尿中枢蓝斑及其周围DA含量的动态变化.结果电针可降低L-多巴引起的膀胱机能亢进大鼠的排尿频率和膀胱内压;降低蓝斑处DA含量.结论电针对L-多巴诱发的大鼠膀胱机能亢进具有良好调整作用.其机理与电针抑制投射到蓝斑的DA能神经元DA释放,导致DA与蓝斑排尿中枢NA能神经元的DA受体结合减少有关.Objective To investigate the neurobiological mechanism of acupuncture inhibiting L-dopa-induced hyperactivity of rat’s bladder by observing changes in micturition frequency, bladder pressure and neurotransmitter dopamine(DA) content at pontine micturition center. Methods A rat model of bladder hyperactivity was made by intraperitoneal injection of carbidopa for inhibiting peripheral dopa decarboxylase, followed by intraperitoneal injection of L-dopa, which was decarboxylate into dopamine in the center to excite pontine micturition center. When the rat was conscious and moved freely, the effect of electroacupuncture on micturition frequency and bladder pressure was dynamically observed by continuous cystometry. Microdialysis and electrochemical high-performance liquid chromatography were used to observe and analyze a dynamic change after electroacupuncture in DA content in and around pontine micturition center locus coeruleus. Results Electroacupuncture could reduce micturition frequency and bladder pressure, and DA content of locus coeruleus in rats with L-dopa-induced hyperactivity. Conclusion Electroacupuncture has an inhibitive effect on L-dopa-induced hyperactivity of rat’s bladder. The mechanism is related to electroacupuncture inhibiting the release of DA form dopaminergic neurons projecting to locus ceruleus to reduce the binding of DA to dopaminergic receptors on the noradrenergic neurons in micturition center locus coeruleus.
