摘  要:目的:建立测定人血浆中雷贝拉唑钠的反相高效液相色谱法。方法:采用 Dikma Diamonsil C_(18)分析柱(250mm×4.6mm,5 μm),以0.1 mol·L^(-1)醋酸铵缓冲液(用三乙胺调至 pH=7.5)-乙腈-甲醇(64:31:5)为流动相,流速1.2 mL·min^(-1),检测波长290 nm,柱温40℃。以奥美拉唑作内标,血样用二氯甲烷1次提取。结果:本方法线性范围为5-1600 ng·mL^(-1),r=0.9999,方法回收率在93.26%-99.72%之间,日内和日间 RSD 均小于4%。结论:本方法灵敏、准确,重现性好,适用于雷贝拉唑钠的药代动力学研究。Objective:An RP-HPLC method was developed for the determination of rabeprazole sodium in human plasma.Method:The chromatographic separation was carried out on Dikma Diamonsil C_(18) column (200 mm×4.6 mm,5μm)with the mobile phase consisting of 0.1 mol·L^(-1) ammonium acetate buffer( adjusted pH to 7.5 with triethylamine)-acetonitrile-methanol(64:31:5),flow rate at 1.2 mL·min^(-1).Detection wavelength was set at 290 nm and column temperature was 40℃.Omeprazole was used as the internal standard and plasma samples were extracted by dichloromethane.Results:The linear range was 5-1600 ng·mL^(-1),r=0.9999.The method recovery was within 93.26%-99.72%.Within-day and between-day RSD were less than 4%.Conclusion:The method is sensitive,specific and reproducible for the determination of rabeprazole sodium concentration in human plasma.It is very suitable for pharmacokinetics study of rabeprazole sodium.
