摘  要:目的观察益肾降浊汤对高脂血症小鼠脑缺血再灌注后海马组织琥珀酸脱氢酶(SDH)、细胞色素氧化酶(CCO)活性以及神经细胞内线粒体ATP生成速率的影响。方法在造成小鼠高脂血症基础上,制备脑缺血再灌注模型。采用差速离心法提取脑海马组织线粒体,用生化法测定SDH、CCO活性;用高效液相色谱法测定ATP生成速率。结果模型组小鼠海马组织SDH活性在术后第1、7、15、30d均显著低于正常对照组(P<0.01),且随着时间的延长,酶活性一直处于低水平;CCO活力与SDH活性的变化趋势相同;线粒体ATP生成速率也在各时间点显著低于同期正常对照组(P<0.01)。用益肾降浊汤治疗后,SDH、CCO活性和线粒体ATP生成速率在各时间点较模型组明显升高(P<0.05,<0.01)。结论益肾降浊汤可抑制脑缺血再灌注造成的SDH、CCO活性降低,提高线粒体ATP生成速率,改善细胞的能量代谢,保护神经元。Objective To investigate the effects of YSJZ decoction on succinodehydrogenase (SDH),Cytochrome C oxidase (CCO) activities and velocity of mitochondrial ATP synthesis induced by cerebral ischemia and reperfusion in the hippocampus of hyperlipemia mice.Methods The model was established by repeated ischemia and reperfusion in hyperlipemia mice.The mitochondria were prepared by centrifugation,the activities of SDH and CCO were measured by biochemical methods and the velocity of mitochondrial ATP synthesis was determined by HPLC.Results The activities of SDH and CCO in hippocampus of model groups on 1,7,15 and 30d after operation were obviously lower than that of normal control groups (P<0.01) and decreased continually with the prolongation of time.The velocity of mitochondrial ATP synthesis in hippocampus of model groups were significantly lower than that of normal control groups at every time point (P<0.01).The activities of SDH and CCO in hippocampus and the mitochondrial ATP synthesis velocity of treatment groups at every time point after operation were higher than that of model groups(P<0.05,<0.01).Conclusion The YSJZ decoction exerts its protective effects to neuronal after cerebral ischemia and reperfusion via inhibiting the decrease of SDH and CCO activities,increasing the mitochondrial ATP synthesis velocity and improving the disorder of energy metabolism in hyperlipemia mice.
