654-2对大鼠肝缺血再灌注损伤的影响探讨

摘　　要：目的探讨654-2对肝缺血再灌注损伤(I-R)的保护作用及对P-selectin蛋白表达的影响机制。方法20只Wistar大鼠随机分为I-R组和654-2组,I-R模型由夹闭门静脉左支45min再开放3h制备,比较两组动物血清ALT、GSH和肝组织P-selectin蛋白水平变化。结果654-2组ALT、GSH指标显著低于I-R组(P<0.01),654-2可显著改善肝细胞镜下损伤并减少肝组织P-selectin蛋白的表达。结论654-2可通过下调肝细胞P-selectin蛋白的表达而对肝缺血再灌注损伤(I-R)有保护作用。Objective To study the protective effects of 654-2 on hepatic ischemia-reperfusion injury (I-R) and P-selectin protein expression in rats. Methods 20 male Wistar rats were randomly assigned into the control group(I-R group) and preconditioning group(654-2 group).After 45 minutes closure of the animals'left portal veins, blood sample、ALT、GSH、 hepatic tissue regular-dye and P-selectin dye were studied 3 hours after the re-open. Results Compared with I-R group,654-2 group ALT、GSH were significantly decreased(P<0.01).Microscopic P-selectin dye were singnificantly decreased due to effect of 654-2.Conclusion 654-2 has protective effectiveness on hepatic ischemia-reperfusion injury by dreasing the expression of hepatic P-selectin.
