摘 要:观察了大骨节病(KBD)患者血清对培养软骨细胞蛋白多糖(PG)合成代谢和分解代谢的影响。实验结果表明:(1)在10%KBD血清存在下培养的静止软骨细胞其PG合成明显低于非病区健康对照组(P<0.01);(2)培养液中添加KBD血清(5%)培养的肥大软骨细胞其释放到培养液中的PG含量显著高于非病区健康对照组(P<0.01),其中低分子量PC的含量增加,缺乏透明质酸结合区,不能与透明质酸结合形成PG聚合体,以上结果指出KBD患者血清中存在干扰PG代谢的物质。Effect of serum from Kaschin-Beck disease (KBD)patients on synthesis and degradation of proteoglycan in chondrocyte cultures was examined. The results shown that (1)when compared to control culture, KBD serum (10%)caused remarkably a decrease of the synthesis of proteoglycan in resting chondrocyte cultures (P<0.01);(2)In the presence of 5% of KBD serum,the content of proteoglycan released into the medium was greater than that of control in hypertrophic chondrocyte culture(P<0.01). Many of proteoglycan monomer released into medium demonstrated that greatly diminished hydrodynamic size,and capacity for hyaluronic acid binding,which had non-functional or missing hyaluronic acid region. These results indicated that there were factors interrupting to metabolism of proteoglycan in KBD serum.
