摘 要:目的观察新的二氢吡啶类钙拮抗剂2,6二甲基1,4(3硝基苯基)1,4二氢3,5吡啶二羧酸3甲酯5正戊酯(MN9202)对家兔血流动力学的影响。方法家兔经乌拉坦氯醛糖混合麻药麻醉,进行左心室、股动脉和下腔静脉插管,同时开胸用电磁流量计测定主动脉根部血流量作为心排出量指标。采用计算机联机实时分析方法,观察MN9202的血流动力学效应。结果间隔10min,按累积剂量方法分别静脉注射MN92021、5、10μg/kg,随着给药剂量的增加,动物的血压、总外周阻力显著下降,与此同时,心排出量(CO)显著增加,心脏泵血功能改善,具有明显的剂量依赖关系。结论MN9202具有显著的的血流动力学效应,不仅作用强,而且持续时间长。AIM: To study the hemodynamic effects of a novel dihydropyidine calcium channel blocker, methyl pentyl 1,4-dihydro-2,6-dimethy l, 4-(3-nitrophenyl)-3,5-pyridenedicarboxylate (MN9202), on anaesthetic rabbits. METHODS: Rabbits were anaesthetized with sodium pentobarbitone. Three polyethylene catheters were inserted into the left ventricular cavity, femoral artery and inferior vena cava, respectively. Blood flow of aortic artery was detected by electric-magnetic flowmeter. Hemodynamic indexes of anaesthetic rabbits were collected and the effects of MN 9202 were analyzed by an affiliated computer in real time. RESULTS: MN9202 was given in an accumulated doses of 1,5 and 10 μg/kg at 10 min intervals. MN9202 exerted significant hemodynamic effects in a dose-dependent manner. With an increasing size of MN9202 dosage, blood preasure and total peripheral resistance of rabbits declined. In the meantime, cardiac output was increased, affecting the improvement of heart function. CONCLUSION: MN9202 has strong and prolonged effects on the hemodynamic indexes of the rabbits .
