乐果对U87细胞HSP70表达及AChE活性的影响

摘　　要：[目的]研究乐果对神经胶质瘤细胞U87热应激蛋白70 (HSP70 )表达的影响规律,比较HSP70与乙酰胆碱酯酶(AChE)对乐果的敏感性。[方法]以不同浓度乐果(10 0、10、1、0 .1、0 .0 1、0 μmol/L)对体外培养的U87细胞分别进行2、4、8、16和2 4h染毒,再分别用Western blot法和DTNB法检测各组细胞HSP70表达和AChE活性的变化,比较这两个指标变化的幅度以及引起此变化所需乐果浓度和染毒时间的差异。[结果]乐果染毒2h即可引起HSP70表达的显著增高(P <0 .0 1) ,这一变化在染毒4h时出现于各剂量组,并可持续至染毒16h。在染毒2h和4h ,HSP70表达的变化与乐果剂量之间存在相关关系(Pearson相关系数分别为0 .916和0 .989,P <0 .0 1)。AChE活性的显著降低出现在染毒4h ,且仅出现在剂量较高的3个组,变化幅度也较小。[结论]乐果可以在相对较短时间内以较低剂量引起U87细胞HSP70表达的显著变化,并存在一定的剂量 反应关系。HSP70比AChE对乐果更为敏感。Objective] To investigate the effects of dimethoate on Heat Shock Protein 70(HSP70)expression in glioma cells and compare the sensitivity to dimethoate between HSP70 and acetylcholinesterase(AChE). [Methods] The U87 cells were treated with different concentrations of dimethoate(100,10,1,0.1,0.01,0 μmol/L)for 2 h,4 h,8 h,16 h and 24 h respectively.Then the method of Western-blot and DTNB were used respectively to test the expression of HSP70 and the AChE activity,comparing the extent of the changes in relation to dosage and treatment time. [Results] Dimethoate treatment can increase the expression of HSP70 significantly in the 2-hour treatment(P<0.01). This change can be seen at all concentrations in the 4-hour treatment and can last for 16 hours.In the 2-hour and 4-hour treatment,the change of HSP70 expression has significant relationship with the concentrations of dimethoate,while the significant decrease of AChE activity appeared in the 4-hour treatment and only in the three higher doses with a lower extent. [Conclusion] Dimethoate can cause the change of HSP70 expression in U87 cells for shorter time and at lower doses .Meanwhile,there is a dose-response relationship.HSP70 may be more sensitive to dimethoate than AChE.
