摘 要:目的:通过研究人牙周韧带细胞(PDLCs)与聚羟基乙酸(PGA)无纺网共培养物在裸鼠体内的生长,探讨PGA无纺网作为支架应用于牙周组织工程的可能性。方法:将共培养7d的人PDLCs与PGA无纺网复合物接种于BALB/C裸小鼠一侧背部皮下,另一侧植入空白PGA为对照。分别于1、2、3、4周取材进行大体及HE和Masson's染色及I型胶原免疫组织化学观察,并进行新生毛细血管记数。结果:随着PGA无纺网在体内的逐渐降解,PDLCs同步增殖,分泌胞外基质。体内2周时即可见实验侧植入物血管化良好并有胶原束的形成,Ⅰ型胶原表达阳性。结论:PGA无纺网具有良好的生物相容性和生物安全性,并易于新生血管的形成,作为牙周组织工程的细胞支架有良好的应用前景。Objective: The aim of this study was to investigate the suitability of 3D polyglycolic acid (PGA )mesh biomaterials as scaffolds for human periodontal ligament (PDL) cell, using a nude mice animal model. Methods: Human PDLCs were cultured and seeded into 3D PGA non-woven fiber meshes scaffolds for 7 days' incubation in vitro. Then each of the twenty four BALB/C-nude mice was implanted with cell-PGA constructs and PGA mesh as a control subcutaneously on the back bilaterally. The mice were sacrificed in batch at 1,2,3 or 4 weeks and the harvests were examined histologically and immunohistochemically. HE staining, Masson's staining and type I collagen immunohistochemical examination were performed. The number of blood vessels present in cell-PGA constructs and control meshes explanted at each time point was counted. Results: The results indicated that the human periodontal ligament cells adhere to PGA scaffolds well and exhibit the excellent matrix secretion ability. Type I collagens could be detected in 2 weeks of implantation by immunohistochemistry and the implants were well-vascularized in vivo. Conclusions: These results suggested that non-woven PGA mesh may serve as a viable scaffold for periodontal tissue engineering.
