摘　　要：研究人永生化支气管上皮BEP2D细胞中,作为Smad蛋白家族的抑制分子,Smad7对TGF-β信号通路中Smad2、Smad3、Smad4核转位的抑制作用。培养BEP2D细胞,瞬时转染Smad7真核表达载体pCISmad7.neo,TGF-β刺激,提取细胞核蛋白及总蛋白,用Westernblot方法比较瞬时转染Smad7基因前后细胞核中Smad2、Smad3、Smad4蛋白表达的差异。结果,Smad3在TGF-b作用下有明显的核转位;转染Smad7后Smad3、Smad4的核转位显著受到抑制。表明在BEP2D细胞中,Smad7对TGF-β/Smads信号通路的拮抗作用主要通过抑制Smad3的活化、Smad3/Smad4异源复合物的形成及核转位,从而拮抗TGF-β对细胞的生长抑制效应。To analyze the effect of Smad7 gene on nuclear translocation of Smad2/3/4 in the immortalized human bronchial epithelial cells BEP2D. Transient transfect full-length Smad7 cDNA construct pCISmad7.neo into BEP2D cells, cell nuclear protein and total protein were extracted, then perform Western blot to examine the expression level of Smad2, Smad3 and Smad4. In BEP2D cells, Smad3 has evident nuclear translocation with stimulated by TGF-β. The nuclear translocation of Smad3 and Smad4 has been inhibited after transfect of pCISmad7.neo. Smad7 negatively regulates TGF-β/Smads signaling through blocking the phosphorylation of Smad3, the formation of Smad3/Smad4 heterogenetic complexes and nuclear translocation.
