摘  要:目的 探讨超声辐照对人肝癌细胞株HepG2 细胞通透性的影响以及声学造影剂声诺维(SonoVue)的增强作用。方法 用连续多普勒超声辐照人肝癌细胞株HepG2,超声发射频率1.9 MHz,软组织热指数(TIS)为0.8,增益设定为80%,输出声强(ⅠSATA)为80.0 mW/cm2,辐照时间均持续60 s。照射时添加或不添加超声造影剂声诺维(SonoVue)。用流式细胞仪检测细胞荧光染色阳性的阳性率。结果 荧光染色阳性细胞的百分率:无超声辐照组或单纯添加SonoVue组分别为(3.06±1.57)%和(1.4±0.04)%,经超声辐照后为(23.72±14.4)%,添加SonoVue辐照后显著增加至(54.78±43.6)%(均P<0.05)。结论 超声辐照能改变HepG2细胞膜通透性,添加超声造影剂后能显著增强这种作用,有希望成为肿瘤分子生物学治疗的介导手段。Objective To explore the role of ultrasound radiation on the permeability of HepG2 cell with/without the intervention of ultrasound contrast SonoVue. Methods HepG2 cell suspension infused with Fluorescent dextran molecules (FD500) was exposed to CW Doppler ultrasound, 1.9 MHz, Tis 0.8, gain 80%,Ⅰ SATA 80.0 mW/cm 2, with/without SonoVue infused. Flow cytometry was used to detect the fluorescent rate. Results The uptake rate of fluorescent cells in control groups (pseudo-US exposure group and only with micro-bubble contrast agent group) were (3.06±1.57)% and ( 1.4 ±0.04)% respectively. Significant uptake increases were found in the US exposure group ([23.72±14.4]%) and US with micro-bubble contrast group ([54.78±43.6]%). Conclusion Diagnostic ultrasound destruction of contrast-agent microbubbles can enhance the permeability of HepG2 cell membrane, which contributes to the delivery of large molecules. It maybe become a promising biomolecular therapeutic method on oncology.
