摘 要:目的观察Wistar大鼠胚胎神经干细胞及其分化细胞的端粒酶活性,探讨神经干细胞维持其生物学特性的分子生物学基础及神经干细胞分化调控的内在机制.方法显微镜下分离大鼠胚胎室管膜下区细胞,在含生长因子的神经干细胞完全培养液中培养形成神经干细胞,然后在不含生长因子而含10%血清培养液中分化14~16 d,应用改良的端粒重复序列扩增法(TRAP)研究神经干细胞及其分化细胞的端粒酶活性.结果 Wistar大鼠胚胎神经干细胞表达较强的端粒酶活性,而分化后的细胞则不再表达端粒酶活性.结论端粒酶活性的改变可能是胚胎神经干细胞诱导分化的机制.随着端粒酶活性的逐渐下降,胚胎神经干细胞随之分化为具有一定生物学特性的细胞.Objective To study the telomerase activities of fetal rat neural stem cells and their differentiated cells,the molecular biological foundation of the neural stem cells maintaining their biological features and the mechanism of differentiation modulation of the neural stem cells.Methods Neural stem cells cultured from the E13 Wistar rat SVZ cells were cultured in the media containing B27,EGF and bFGF without blood serum.Then neural stem cells were induced to differentiate with 10% blood serum in the absence of growth factor for 14 to 16 days.Telomeric repeat amplification (TRAP) was used to study the telomeric activities of neural stem cells and differentiated cells.Results The fetal neural stem cells showed strong activity of telomerase.The telomerase activities were imprisoned in differentiated progenies of neural stem cells.Conclusion The process of differentiation of fetal neural stem cells may rely on the gradual devitalization of telomerase activity.Neural stem cells differentiate with the descending of telomeric activities.
