摘 要:目的:研究环氧氯丙烷(epoxychloropropane,EC)处理对生物瓣膜在体外钙沉积实验中的防钙化作用以及体外钙沉积实验和动物皮下包埋实验钙化模型的比较,探究EC处理生物瓣膜防钙化的机制。方法:实验于2003-03/2004-04在解放军第四军医大学西京医院全军心血管外科研究所实验室完成。采用体外钙沉积和大鼠皮下包埋两种钙化模型,对新鲜猪主动脉瓣膜(新鲜瓣膜组)、戊二醛处理(戊二醛处理组)、EC处理(EC处理组)以及戊二醛、EC联合处理(戊二醛+EC处理组)的猪主动脉瓣膜进行体内、体外钙化研究,体外钙沉积实验中采用1.5倍血浆离子浓度的液体进行模拟钙沉积实验,体内实验动物为SD大鼠24只,随机分为3组(2周组,4周组,6周组),每组8只,分别于脊柱两侧头端尾端4个部位包埋各组瓣膜各1例,分2,4,6周分别取出瓣膜,通过原子吸收法测定钙含量。结果:在体外钙沉积实验中各组瓣膜的钙化程度在第3,7,14天时递增,最终在第21天EC处理组和戊二醛处理组瓣膜的钙含量明显高于新鲜瓣膜组(t=5.662,16.436,P<0.05);EC处理组和戊二醛+EC处理组瓣膜的钙含量明显低于戊二醛处理组P<0.05)。大鼠皮下包(埋实验第6周戊二醛处理组和戊二醛+EC处理组瓣膜的钙化含量犤(380.8±67.3),(84.0±9.4)μmol/g犦明显高或低于新鲜瓣膜组犤(173.2±44.7)AIM: To study the anticalcification of epoxy chloropropane (EC) treatment on biovalve in vitro, and compare the in vitro experiment of calcium deposition with animal subcutaneous embedding experimental calcification model, and investigate the mechanism of EC treatment against calcification of biovalve.METHODS: The experiments were done in the laboratory of the Department of Cardiovascular Surgery, Institute of Cardiology, Xijing Hospital of the Fourth Military Medical University of Chinese PLA from March 2003 to April 2004. Two kinds of calcification models of calcium deposition and subdermal embedding rats were used in this study.The porcine aortic valves were divided into four groups: fresh porcine valve group,glutaraldehyde(GA) treated valve group (GA group), EC treated valve group (EC group) and GA+ EC treated group (GA+ EC group) for the study of in vivo and in vitro calcification. In the experiment of in vitro calcium deposition, liquid containing 1.5 times plasma ion was used to imitate calcium deposition.In the in vivo experiment, 24 SD rats were randomly divided into 3 groups with 8 rats in each group: 2,4 and 6 weeks groups. The valve of rats was embedded at the tip and end of bilateral spine respectively, and one rat was used for each site, and then the valves were taken out at 2,4 and 6 weeks respectively, and the calcium content was measured with atomic absorption spectroscopy.RESULTS: In the calcium deposit experiment in vitro, the degree of calcification in each group was increased gradually at the 3rd, 7th and 14th days, finally at the 21st day,the calcium contents were obviously higher in the EC and GA groups than in the fresh valve group (t=5.662,16.436,P< 0.05), and those were markedly lower in the EC group and GA+ EC group than in the GA group (P< 0.05). At the 6th week of the rat subdermal embedding test, the calcium content in the fresh valve group [(173.2± 44.7) μ mol/g] was significantly lower than that in the GA group [(380.8± 67.3) μ mol/g], but remarkably higher than that in the GA
