摘  要:目的:应用异体许旺细胞结合聚乳酸-聚羟基乙酸共聚物管及细胞外基质凝胶构建组织工程人工神经复合体,并观察移植许旺细胞的分布及存活情况。方法:实验于2003-03/06在第三军医大学大坪医院野战外科研究所,创伤,烧伤与复合伤国家重点实验室完成,以组织工程人工神经复合体坐骨神经缺损桥接动物模型,应用Hoechst33342荧光预标记技术观察移植许旺细胞的分布及存活情况。结果:荧光显微镜观察显示,异体移植的许旺细胞呈较均匀的散在分布,其存活时间可达4周左右。结论:实验条件下,异体许旺细胞移植在缺乏免疫抑制治疗的情况下可在受体组织中存活一定时间,这一结果为下步实验应用异体许旺细胞构建组织工程人工神经复合体治疗神经缺损并可能发挥作用提供了实验基础。AIM:To observe the viability and distribution of allogenic Schwann cells in ti ssue engineered artificial nerve complex constructed with poly(DL lactide co glycolide) conduit and extracellular matrix when bridging sciatic nerve defect in rats. METHODS:The experiment was conducted in the Research Institute of Field Surger y,Daping Hospital,Third Military Medical University of Chinese PLA,and State Key Laboratory for Trauma,Burn and Combined Injury,from March to June 2003.Based on the animal models of tissue engineered artificial nerve complex when bridging sciatic nerve defect,allogenic Schwann cells were pre labeled with Hoechst33342 to observe their distribution and viability by fluorescence microscopy. RESULTS:As shown by fluorescence microscopy,the transplanted Scwann cells dist ributed evenly,and could survive for about 4 weeks after implantation. CONCLUSION:Under the condition of our experiment,the implanted allogenic Scwan n cells can survive for a certain time in the recipient tissues in the absence o f immunosuppressive therapy,which will provide an experimental basis for the fur ther study on the construction of tissue engineered artificial nerve with allog enic Scwann cells for nerve defect and the possible roles.
