摘  要:背景:创伤修复是一个在时间和空间上受修复因子调控的复杂生物学过程,其中细胞因子在创伤修复中的活跃表达,已成为推断损伤时间的新的参考参数,皮肤创伤愈合过程中的表达、分布与损伤时间的关系目前尚不十分清楚。目的:了解细胞因子在小鼠皮肤创伤愈合过程中的表达、分布与损伤时间的关系。设计:以实验动物为对象,随机对照的验证性研究。单位:第一军医大学病理教研室暨全军分子肿瘤重点实验室。材料:实验于2003-07/09在全军分子肿瘤重点实验室完成。清洁级昆明鼠65只,雌雄不拘,体质量25~28g。干预:随机分为实验组和对照组,实验组按生前不同造创时间分为12组(伤后15,30min,1,3,6,12,24,48,72,96h,1,2周),每组5只动物。5只未造创正常小鼠作为对照组。主要观察指标:不同损伤时间皮肤炎性细胞、修复细胞血管内皮细胞生长因子(VEGF)、转化生长因子β1(TGF-β1)、碱性成纤维细胞生长因子(bFGF)表达免疫组化染色的强度及分布。结果:VEGF在表皮及皮脂腺呈持续性阳性表达,TGF-β1、bFGF于伤后3h在表皮表达增强,并分别持续至伤后12h和48h。三种细胞因子在炎性细胞和修复细胞中的表达变化规律相似,伤后6h主要在部分中性粒细胞表达,12h后则主要在巨噬细胞和成纤维细胞。阳性细胞数量随损伤时间的延长而增多。BACKGROUND:Wound healing is a complex biological process regulated temporally and spatially by many repair factors.Studies have showed the predominant express ion of cytokines in wound healing process,which is a kind of molecular vital rea ction,and may be a new reference index in wound age estimation. But the precise relation of the expressions of cytokines and distribution to wound age is not cl ear. OBJECTIVE:To probe into the relation of the expressions of cytokines and distr ibution to wound age in the healing process of the mouse skin wounds. DESIGN:A randomized controlled confirmation study based on the experimental an imals SETTING:Department of Pathology of the First Military University of Chinese PL A,which is a key laboratory of molecule oncology of Chinese PLA. MATERIALS:The study was completed in the Key Laboratory of Molecule Oncdogy of Chinese PLA from July to September in 2003.Sixty five Kunming mice of either g ender,weighing 25-28 g,were involved. INTERVENTIONS:The mice were randomly divided into the experimental and the con trol group.The mice in the experimental group were divided into 12 subgroups acc ording to the different times of trauma.The mice were killed at the following ti me intervals:15 minutes,30 minutes,1 hour,3 hours,6 hours,12 hours,24 hours,48 h ours,72 hours,96 hours,1 week or 2 weeks after wound and the wounded skin was ex cised(n=5 each).As a control,5 mice without wounding were examined. MAIN OUTCOME MEASURES:The amount and distribution of staining positive inflamm ation and repair cells in skin wound tissues. RESULTS:Persistent positive expression of vascular endothelial growth factor(V EGF) was seen in the epidermal cells and sebaceous gland cells.The expressions o f transforming growth factor β1(TGF β1) and basic fibroblast growth factor ( bFGF)in the epidermal cells were increased at 3 hours after incision and lasted till 12 hours and 48 hours respectively.There was a similar distribution in the positive amount of inflammation and repair cells among VEGF,TGF β1 and b
