摘  要:目的研究mPEG阻断体液免疫、细胞免疫的机理。方法用浓度梯度法修饰淋巴细胞表面抗原,通过微量淋巴细胞毒试验及淋巴细胞转化试验检测mPEG对HLA的遮蔽效果;通过淋巴细胞表面CD分子及单向混合淋巴细胞培养检测对细胞免疫的阻断;体外保存淋巴细胞检测其寿命。结果修饰后淋巴细胞表面HLA-I类抗原与其相应抗体的微量淋巴细胞毒试验为阴性且淋巴细胞相对转化指数明显降低;修饰后淋巴细胞表面CD分子荧光强度及淋巴细胞转化率明显降低;修饰前后淋巴细胞寿命无明显改变。结论用mPEG对淋巴细胞表面抗原进行修饰后,可以阻断体液免疫及细胞免疫而不影响淋巴细胞的存活。Objective To study the mechanism of humoral and cellular immunity blockage induced by methoxypolyethylene glycol (mPEG). Methods The antigens on the surface of lymphocytes were modified by mPEG concentration gradient. The effect of the modification on HLA was detected by microlymphocytotoxicity test and lymphocyte transformation test. The blockage of the cellular immunity was detected by CD expression and one-way mixed lymphocyte culture (MCL). The lifetime of lymphocytes was evaluated by the culture in vitro at 22℃. Results The results of microlymphocytotoxicity test of the lymphocytes disposed by mPEG-benzotriazole carbonate (BTC) were negative and the relative response index of lymphocyte was decreased. After the modification, the fluorescence intensity of CD and the lymphocyte transformation efficiency of one-way MCL were decreased obviously. No significant difference of lifetime was found in lymphocytes before and after modification. Conclusion The humoral and cellular immunity could be blocked by mPEG while the lifetime of lymphocytes are not influenced.
