摘 要:目的:探讨天麻超微粉对大鼠脑缺血再灌注损伤的神经保护作用。方法:采用大鼠脑缺血2小时再灌注24小时的局灶性缺血再灌注模型。用 HE 染色、2,3,5-三苯基氯化四氮唑(TTC)染色、免疫组化方法分别观察使用天麻超微粉后脑片的病理改变、脑梗塞的体积和神经细胞凋亡相关蛋白 Bcl-2、Bax 表达的变化。结果:(1)各组脑梗塞体积比较:天麻超微粉能缩小脑梗塞的体积,其低、中、高剂量组与缺血组相比差异均有显著性(P<0.05或 P<0.01);(2)HE 染色结果:天麻超微粉低、中、高剂量组缺血半球纹状体残存细胞百分率与缺血组相比差异均有显著性(P<0.01);(3)各组 Bcl-2/Bax 比值比较:缺血组与假手术组相比差异有显著性(P<0.01);天麻超微粉低、中、高剂量组与缺血组相比差异均有显著性(P<0.01)。结论:天麻超微粉具有抗脑缺血再灌注损伤的神经保护作用,其作用可能与调控凋亡相关蛋白 Bcl-2、Bax 的表达有关。Objective:To explore protective action of ultramicro-powder of Rhizoma Gastrodiae on nerves in the rat of Cerebral ischemia- reperfusion injury.Methods:The local ischemia-reperfusion injury at model was established by cerebral ischemia for 2 hours and reperfusion for 24 hours.HE staining,2,3,5-triphenytetrazolium(TTC)staining,immunohistochemical methods were respectively used to investigate pathological changes of brain slices,the size of cerebral infarction and nervous cell apoptotic relative proteins Bcl-2,Bax expression after treatment with ultramicro-powder of Rhizoma Gastrodiae.Results:(1)Ultramicro-powder of Rhizoma Gastrodiae could reduce the size of cerebral infarction.When the low,middle and high dose groups were compared with the ischemia group there were significant differences(P<0.05 or P<0.01);(2)There were significant differences in the percentage of survival cells of the striatum of ischemic hemisphere in ultramicro-powder of Rhizoma Gastrodiae low,middle and high dose groups when compared with the ischemic group(P<0.01);(3)There was a very significant difference in Bcl-2/Bax ratio between the ischemia group and the sham operation group(P<0.01);and between the Rhizoma Gastrodiae ultramicro-powder low,middle and high dose groups compared and the ischemia group(P<0.01).Conclusion:Ultramicro-powder of Rhizoma Gastrodiae has protective action on the nerve in the ischemia-reperfusion injury,which is possibly related with apoptosis-relative proteins Bcl-2 and Bax expression.
