摘  要:目的 研究粘着斑激酶在不同年龄大鼠各组织中的分布以及在血管再狭窄和动脉粥样硬化血管中的表达。方法 应用Westernblot技术研究粘着斑激酶在不同年龄大鼠各组织中的分布以及在血管内皮剥脱术后再狭窄和动脉粥样硬化血管中的表达。结果 粘着斑激酶在1月龄幼年大鼠各组织中的表达较成年大鼠明显升高;在血管内皮剥脱术后3,7,14天粘着斑激酶表达逐渐升高,2 1天有所下降,但仍高于对照组;粘着斑激酶在动脉粥样硬化血管中的表达较对照组升高。结论 粘着斑激酶可能参与胚胎发育后各组织的发育调节;粘着斑激酶可能参与血管成形术后再狭窄及动脉硬化的病理过程,粘着斑激酶表达的上调可能在血管成形术后再狭窄及动脉硬化的发病过程中起重要作用。Objective To study the expression of focal adhesion kinase (FAK) in tissues of rats at different ages and in restenosed or arteriosclerosti c(AS) vessels.Methods Vascular de-endothelialization and feedi ng with fat-rich diet and high dose of vitamin D3 were performed to establish animal models of restenosis and AS.Western blot was employed to detect level of FAK protein by using polyclone antibody against FAK.Results The expression of FAK in tissues of one-month old rat was much higher than those in older ones. The amount of FAK protein significantly increased 3,7,and 14 days after de-endothelialization,compared with control group.During the ob servation time, the amount of FAK protein reached the peak on day 7, and began t o decrease on day 14. The expression of FAK protein marginally descended on day 21 but was still higher than basal level in control group.The expression of FA K in AS aorta was higher than that in control rats.Conclusions FAK may be involved in the regulation of development of different tissues in you ng rats a nd the pathogenesis of restenosis and AS.Up-regulated expression of FAK may be one of the critical mechanisms in causing intima thickening after endotheli um injury and arteriosclerosis.
