摘  要:目的:探讨抗代谢药物丝裂霉素C(mitomycinC,MMC)对实验性椎板切除术后硬膜外瘢痕组织中羟脯氨酸(HOP)含量的影响。方法:手术切除SD大鼠第一腰椎椎板,造成0.2cm×0.5cm硬脊膜裸露区,按所用MMC的不同浓度随机分为4组即高剂量组0.1mg/ml、中剂量组0.05mg/ml、低剂量组0.01mg/ml及生理盐水对照组,每组10只。术后第4周末处死动物,测定其硬膜外瘢痕组织HOP含量,并按Rydell法评定粘连程度。结果:MMC各剂量组HOP含量均较对照组降低,高剂量组HOP含量明显低于对照组,且具有显著性差异(P<0.05);和对照组比较,中剂量组和低剂量组也显示了降低趋势,但其差别未能达到显著性水平(P>0.05);从MMC处理组来看,随着剂量的增加,HOP的含量具有降低趋势,具有剂量-效应关系。大体观察也发现高剂量组粘连明显减轻。结论:丝裂霉素C可抑制瘢痕组织中HOP含量,减少SD大鼠模型硬膜外瘢痕组织胶原的形成。Objective: To study the effect of mitomycin C, an anti-metabolic drug, on content of Hydroxyproline in the epidural scar tissue of experimental post-laminectomy. Methods: 0.2 cm×0.5 cm dura mater uncovered area laminectomy was performed at L_1 in sixteen SD rats, which were divided into four groups randomly by various mitomycin C concentrations, They were Group A_1(0.1 mg/ml mitomycin C), Group A_2(0.05 mg/ml Mitomycin C), Group A_3(0.01 mg/ml mitomycin C), Group C(normal saline as control). The animals were sacrificed four weeks after operation. The specimens were prepared for determination of the content of Hydroxyproline at scar tissue and the degree of scar adhesion according to Rydell method. Results: The content of Hydroxyproline in every mitomycin C-treated group was decreased while compared to that of Group C.The content of Hydroxyproline of Group A_1 was lower than that of Group C(P<0.05),The content of Hydroxyproline between Group A_2 and Group A_3 had insignificant difference(P>0.05), however, they both were lower than the content of Group C(P<0.05). The contents of Hydroxyproline in Group C. Group A_(3.) Group A_(2.) Group A_1 were decreased with the trend of increasing concentration. Conclusion: Mitomycin C inhabited an increase of Hydroxyproline in the scar tissue and reduced epidural scar tissue collagen formation of SD rat models.
