摘 要:用亚MIC连续递增式传代培养法获得了阴沟杆菌1029对三种氟喹诺酮类药物(Fluoroqu-infones,FQs)稳定的高度耐药株(MIC提高≥128倍),观察到FQs之间存在完全交叉耐药,且此耐药株对氯霉素、四环素、头孢噻肟和氨曲南也呈耐药现象(MIC提高≥8倍),但对庆大霉素和哌拉西林的敏感性无明显改变。进一步研究结果提示:膜通透性降低致使药物入胞量减少是阴沟杆菌多重交叉耐药性产生的重要原因,它可能与菌体二种外膜蛋白(分子量为37和135千道尔顿)减少或缺失有关。Stable and high level fluoroquinolones(FQs)-resistant strains of Enterobacter cloacae 1029 were obtained with serial passage onagar containing subinhibitory concentrations of indicated agent(Norfloxa-cin, Enoxacin,or Pefloxacin),It was found that cross resistance existed among FQs,Chloramphenicol(CM),Tetracycline(TC),Aztreonam(AZ),and Cefotaxime(CTX),but the multi resistant strain was still sensitive to Pipracillin(PIP)and Gentamicin(GM),The intake of FQs by bacteria was tested.The results suggested that one reason of bacterial resistance to FQs was the reduction of its menibrane permeability,The components of bacterial membrane proteins were also analyzed by SDS-PAGE method.Based on documents published,it was suggested that the outer membrane proteins(37KD and 35 KD)were porins of Enterobacter cloacae 1029,their deficiency or decrease in quantity was related with multi-resistance of the bacteria.
