氯硝安定血浓度测定的高效液相色谱法

摘　　要：作者在国外工作的基础上,首次在国内建立了用高效液相测定氯硝安定血浓度的方法.该方法以氟硝安定为内标,用异丙醚提取,氮气吹干,流动相(乙睛:0.1mol/1,乙酸钠缓冲液为40:60/v:v)重溶进样.色谱条件:紫外检测波长为 254nm,柱压 10.9MPa,流动相流速1.0ml/min,ATTN为2.回收率为993-101.1%,日内差异为1.8-2.5%,日间变异系数为0.9-6.6%.最小检测限为7μg/l.该方法工作曲线稳定,回收率理想,特异性高,操作步骤简便,已应用于儿科临床.Clonazepam(CZP) is widely used in therapy of children with epilepsy due to its efficacy and safety. To further study the pharmacokinetics and pharmacodynamics, we have first developed the method of determinating CZP blood levels by using high performance liquid chromatography. Flunatrizepam is used as a internal standard. The recovery rate of CZP was 99. 3 - 101. 1% ; interassay evaluation was 1. 8-2.5% ; interassay evaluation was 0.9 -6.6%. Other current anticonvulsants did not interfere with CZP peak. It already used into our clinic. Practice suggested this method be accuracy, precise, specific and convenience, it is valuable to use both in clinic and research.
