猪繁殖与呼吸综合征胶体金抗体检测技术的建立和初步应用

摘　　要：用胶体金标记纯化后的猪繁殖与呼吸综合征病毒(PRRSV)基因工程表达抗原,建立一种以微孔滤膜为固相载体,以红色胶体金为标记物的检测PRRSV抗体的胶体金免疫层析法(GICA)。特异性交叉试验证明GICA检测PRRSV抗体有较高的特异性。与其他方法对比检测证实了其敏感性。生产了一批免疫胶体金试纸条,检测36份临床样本,其中30份经免疫胶体金试纸条及ELISA、免疫荧光检测均为阳性。猪繁殖与呼吸综合征免疫胶体金抗体检测技术的建立为检测猪繁殖与呼吸综合征病毒(PRRSV)抗体提供了一种快速简便的方法。A dot immuno_gold filtration assay for porcine reproductive and respiratory syndrome virus(PRRS) was developed.The purified PRRS genetic engeering expressed antigen was labelled by colloidal gold.The results of specificity cross experiment indicated that detecting the PRRSV by GICA was of high specificity.And its sensitivity was improved through comparson with other methods.A batch of colloidal gold test papers was manufactured and 150 clinical samples were detected,30 of which were positive by both ELISA and immuno_fluorescence.The development of this method provide a rapid and convenient means to detect the PRRSV antibody.
