摘  要:目的 建立大鼠异基因骨髓移植模型,受体大鼠按不同的移植物与受体T细胞比例回输供体大鼠骨髓细胞。研究该比例对移植物抗宿主病(GVHD) 的发生及其程度的影响。方法 Wistar大鼠为供鼠,SD大鼠为受鼠,受鼠经全身照射(TBI)+环磷酰胺(CTX)预处理后,分别按不同的移植物与受体T细胞比例(1∶1、2∶1、4∶1)回输供体大鼠骨髓细胞。观察移植后各组受体大鼠生存期及GVHD反应。结果 受体大鼠按4∶1的移植物与受体T细胞比例回输供体大鼠骨髓细胞时,GVHD较严重,而当该比例控制在1∶1 和2∶1时,GVHD程度明显减轻(P<0.05)。结论 移植时将移植物与受体T细胞比例控制在一定范围内,使移植物与受体T细胞保持平衡,既有利于移植物的植入,又可减轻GVHD及其他移植相关并发症的发生。Objective A rat allogeneic bone marrow transplantation model was created with the recipient rats infusing of donor rats’ bone marrow cells at different graft-receptor T cell ratio. The influence of different graft-receptor T cell ratio on the development and the degree of severity of graft-versus-host disease (GVHD) were studied. Methods SD rats were served as the donors and Wistar rats as recipients. The recepients after receiving TBI+CTX regiment were infused donor rats’ bone marrow cells in different graft-receptor T cell ratios (1∶1,2∶1,4∶1), then the survival period and GVHD of each group were observed. Results The degree of GVHD was more severe with this ratio of 4∶1, but milder when it was 1∶1 and 2∶1(P< 0.05). Conclusion During transplantation, the graft-receptor T cell ratio is controlled within certain range, and maintained to keep the balance of grafts and receptor T cells providing a more favorable condition for the transplanted graft, and reducing the occurrence of GVHD with related complications.
