猪生长激素单克隆抗体的制备及其特性

摘　　要：选取7只BALB/c小鼠,以猪生长激素为抗原,采用常用的免疫方法进行免疫,通过细胞融合技术,经筛选克隆,获得了3株分泌猪生长激素单克隆抗体的杂交瘤细胞株,分别命名为1A3、1A5、1B5。亚类鉴定,1A3为IgG1,1A5和1B5为IgG2a。腹水效价依次为1∶3.2×105、1∶3.2×104、1∶6.4×104。亲和力常数依次为1.0×109M-1、0.5×107-1和2.0×108-1。7 BALB/c mice were immunized with porcine growth hormone (pGH),and monoclonal antibodies (mAb) were prepared for hybridoma technique.Three hybridoma cell strains secreting mAbs for porcine growth hormone were obtained,named by 1A3,1A5 and 1B5 separately.By analysis of Ig subclass,the 1A3 was classified as IgG_1,and the 1A5 and 1B5 as IgG_~2a .The ascities fluids titre of 3mAbs was 1∶3.2×105,1∶3.2×104 and 1∶6.4×104 respectively.Comparison of their relative affinity was 1A3>1B5>1A5.
