手性丁基苯酞对暂时性局部脑缺血大鼠脑梗死体积的影响

摘　　要：目的:研究手性丁基苯酞(l-,d-和dl-NBP)对实验性大鼠局部脑缺血的治疗作用。方法:应用插线法造成大鼠暂时性局部脑缺血模型,并通过测定脑梗死体积和行为学评分,比较l-NBP和(d-NBP对局部脑缺血的保护作用。结果:l-NBP组脑梗死体积明显缩小,而且存在剂量效应关系,其中40mg·kg^(-1)组作用最强,缩小梗死体积达80.4%,疗效接近于MK-801组(81.8%)。d-NBP和dl-NBP组作用较弱,并且l-NBP20和40mg·kg^(-1)组的作用明显大于dl-NBP40和80mg·kg^(-1)组。行为学评分与脑梗死体积的结果相吻合。结论:l-NBP抗脑缺血作用较强,d-NBP不仅作用弱,而且阻滞l-NBP的抗脑缺血作用,因此有必要开发l-NBP成为新的治疗急性缺血性脑卒中的药物。Objective:To investigate effects of l-,d-and dl/-3-n-butylphthalide (NBP) on rat focal cerebral ischemia. Methods:Following after orally feeding l-,d-and dl-NBP to rats, respectively, a transient cerebral ischemia model was induced by middle cerebral artery occlusion. Protective effects of the NBPs on ischemic brain injury were assessed by measuring cerebral infarct volume and behavioral scores. Results:The l-NBP showed the dose dependency of effects on the cerebral infarct volume and the behavioral scores. The cerebral infaret volume was reduced by 80.4% when the dose of l-NBP was at 40mg·kg^(-1), which was coincident with effects of MK-801 (81. 8%). The d-and dl-NBP had fewer effects on the cerebral infaret volume and the behavioral scores, compared with the l-NBP.The d-NBP antagonized the neuroprotection of the l-NBP. Conclusion:The l-NBP has a potential protective effects on the focal cerebral ischemia.
