摘 要:目的:研究银杏内酯对外源性血小板活化因子(PAF)加重离体豚鼠心肌缺血再灌注损伤的影响。方法:采用离体豚鼠心脏Langendorff灌流装置,观察银杏内酯(4,8,16mg·L_(-1))对离体豚鼠心肌再灌注心律失常、心肌组织丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)、三磷酸腺苷酸酶(ATPase)以及冠脉流量(CF)的影响。结果:银杏内酯、普萘洛尔有效减少损伤心肌MDA含量及Ca^(2+)-ATP酶活力,增加GSH-PX活力,并且对抗PAF加重的离体豚鼠心肌缺血再灌注心律失常的发生。结论:银杏内酯对抗PAF加重离体豚鼠心肌缺血再灌注损伤的作用与拮抗PAF受体、抑制自由基产生、减少心肌细胞钙超载、防止心律失常的发生有关。Objective:To investigate effects and mechanism of ginkgolides in the protection of myocardial ischemia-reperfusion(MI/R)damage in vitro. Methods: A experimental model of the isolat- ed guinea-pig MI/R damage exacerbated by platelet-activating factor (PAF) was set up to determine if ginkgolides (4,8 and 16mg·L^(-1)) effected activities of glutathione peroxidase (GSH-PX) and ATPase,and the volume of malondialdehyde (MDA) and coronary flow (CF) in myocardial tissue. Results: Ginkgolides significantly reduced the level of MDA in myocardial tissues, increased activities of GSH-PX and Na^+, K^+-ATPase, and inhibited activity of Ca^(2+)-ATPase. Ginkgolides moderately improved the volume of CF in the MI/R damage exacerbated by PAF. Conclusion: Ginkgolides protects occurrence of arrhythmia during the MI/R by antagonizing activities of PAF and generation of free radicals to reduce the overload of calcium in myocardial cells.
