摘 要:本研究在体外将含卵清蛋白 (OVA)的PLGA微囊与小鼠骨髓诱导的树突细胞 (BM DC)相互作用 ,然后用卵清蛋白特异性杂交瘤T细胞检测OVA抗原在BM DC表面的表达 ,结果显示 :PLGA微囊可以在体外显著促进BM DC对OVA抗原的呈递。This paper observed the effects of poly(D. L-lactic-co-glycolic acid) (PLGA) microparticles on antigen presentation of bone marrow-derived dendritic cells in vitro. The DCs were pulsed and chased with OVA encapsulated in PLGA microparticles and then fixed before the addition of OVA-specific DO11·GFP T hybridom cells to analyse the degree of antigen presentation. The significant levels of antigen-presentation could be detected shortly after a pulse with 50ug/ml of encapsulated OVA for 30min-60min. The encapsulation of OVA in PLGA microparticles produced a 100-fold increase in antigen presentation. Our results proved in vitro that encapsulation of antigen inside PLGA microparticles was essential for increased antigen presenting efficiency to be achieved and indicated that the ability of PLGA particles to induce high levels of antigen presentation by DCs may have significant potentials in vaccine adjuvant activity.
