摘 要:目的研究苯那普利、氯沙坦联合治疗对自发性高血压大鼠(SHRs)靶器官的保护作用。方法13月龄SHRs作为实验组,同龄WKY大鼠作为正常对照组,实验组分别予以苯那普利、氯沙坦及苯那普利+氯沙坦联合治疗12w。实验前及实验开始后4、8、12w测量各组血压、24h尿白蛋白排泄;比较实验结束时各组心脏重量、血浆及心脏、肾脏组织AngⅡ、TGFβ1水平。结果苯那普利、氯沙坦及苯那普利+氯沙坦都可以明显降低SHRs血压、24h尿白蛋白排泄,减轻心肌肥厚,但以苯那普利+氯沙坦效果最好。3组中血浆、心肌及肾脏组织中的AngⅡ水平在氯沙坦组升高,在氯沙坦以及苯那普利+氯沙坦组下降。3组中循环和组织的TGFβ1水平都有显著降低,以合用药组降低的程度最大。结论联合应用苯那普利和氯沙坦与单药治疗相比,对于降低SHRs的血压有很好的协同作用,对心脏、肾脏等靶器官有更好的保护作用。Objective To study the protective effect of the combination of benazepril and losartan on the target organ in spontaneous hypertensive rats (SHRs).Methods SHRs aged 13 months were selected as treated groups (with benazepril, losartan, combination of benazepril and losartan respectively for 12 weeks), and WKY rats of the same age as control group. Blood pressure (BP) and ualb excretion for 24 h were measured before and at 4, 8 and 12 weeks after experimentation. Heart weight and AngⅡand TGF-β_1 level in plasma, myocardium and kidney among various groups were compared when the study was finished.Results All benazepril, losartan and benazepril+losartan could lower BP, 24 h ualb excretion, myocardium hypertrophy, especially benazepril+losartan had the best effect. AngⅡlevel in plasma, myocardium and kidney increased in losartan group, decreased in benazepril and combination group. TGF-β_1 level in circulation and tissue in three groups significantly decreased, especially decreased most remarkably in combination group.Conclusions Compared with the other groups, the combination of benazepril and losartan had additive hypotensive effect, and had better protective effect on target organ in SHRs.
