摘  要:目的观察螺内酯对单侧输尿管梗阻(UUO)大鼠肾皮质金属蛋白酶1组织抑制剂(TIMP-1)介导的肾间质纤维化的影响。方法UUO大鼠给予螺内酯20mg·kg-1·d-1灌胃,用免疫组化及RT-PCR的方法检测术后7d、14d、21d的TIMP-1表达量并观察肾脏病理改变。结果与假手术组相比,UUO组及螺内酯组的TIMP-1mRNA和TIMP-1蛋白表达水平均明显增高(P<0.01),且UUO组显著高于螺内酯组(P<0.01)。结论螺内酯可通过下调TIMP-1减轻UUO术后肾组织间质纤维化。Objective To investigate the effect of spironolactone on the expression of TIMP-1 and its mechanism of renal interstitial fibrosis following unilateral ureteral obstruction (UUO)in rat kidney. Methods Fifty- four rats were randomly assigned to shame operation group (shame group), UUO group, spironolactone (20 mg·kg·-1·d-1)treatmeat group after UUO. Immunohistochemistry and RT-PCR were performed to investigate renal pathological changes and examine the expression of TIMP-1 at the 7th, 14th and 21th day in the kidney.Results In comparison with the shame group, the expression of TIMP-1 mRNA and TIMP-1 protein of UUO and spironolactone treatment groups increased significantly (P< 0.01 ),meanwhile spironolactone could significantly decrease the expression of TIMP-1 as compared to UUO group (P<0.01). Conclusion Spironolactone may play a protective role of the kidney by down-regulating TIMP-1 and alleviating renal interstitial fibrosis following UUO.
