维甲酸诱导大鼠肛门直肠畸形胚胎中RARmRNA表达的研究

摘  要:目的探讨胚胎发育中维甲酸诱导的RARmRNA表达异常与肛门直肠畸形发生之间的相关性。方法利用全反维甲酸(ATRA)诱导大鼠产生肛门直肠畸形胚胎,用RTPCR法测定妊娠11、12、13、14、15、16d时大鼠胚胎尾侧部分的RARmRNA表达模式并且与对照组中RARmRNA表达模式进行对比。结果实验组中80%胚胎具有肛门直肠畸形,对照组未见明显畸形。实验组中RARαmRNA表达水平在妊娠11、12、13、15d时低于对照组,表达曲线的高峰出现时间晚于对照组,实验组中RARβmRNA表达水平在妊娠12、14d时低于对照组,而RARγmRNA表达水平在实验组中与对照组中并无明显差异。结论ATRA可诱导胚胎发育过程中RARmRNA异常表达,它可能在妊娠11~13d这一尾侧区域发育最关键时段通过抑制RARα的表达启动并最终导致肛门直肠畸形及其伴发畸形的发生。Objective To explore the relationship between abnormal expression of RAR mRNA and pathogenesis of anorectal malformations (ARMs) induced by retinoic acid. Methods The rat embryos with anorectal malformations were induced by all-trans retinoic acid (ATRA). The expression patterns of RAR mRNA in caudal part of the embryos were determined by RT-PCR on E10 (the tenth gestational day), E11, E12, E13, E14, E15 and E16, respectively. The data from the control group and the ARM group were compared and analyzed. Results In the ATRA treated group, 80% embryos showed anorectal malformations. The expression levels of RARα mRNA in the ATRA treated group were lower than those of control group on E11, E12, E13 and E15. The peak of RARα mRNA expression of the ATRA treated group came later than that of the control group. The expression levels of RARβ mRNA in the ATRA treated group were lower than that of the control group on E12 and E14. The expression levels of RARγ mRNA, however, showed no significantly difference between the control group and the ATRA treated group. Conclusions ARMs induced by ATRA in rats may be mediated by the abnormal expressions of RAR mRNA during the process of embryonic development.
