丙泊酚对缺氧无糖损伤大鼠海马脑片的保护作用

摘  要:目的探讨丙泊酚对缺氧无糖(OGD)损伤脑片的保护作用。方法采用缺氧无糖的人工脑脊液灌流离体大鼠海马脑片模拟缺血损伤,根据给药方式不同分为:OGD前给药组和OGD给药组,均给予1、10和20μmol·L-1的丙泊酚和脂肪乳(溶剂对照)。记录海马脑片顺向群峰电位(OPS)和缺氧损伤电位(HIP)的变化。结果OGD给药组应用10μmol·L-1的丙泊酚,与应用脂肪乳相比OPS消失时间推迟(P<0.05),OPS恢复率升高(P<0.05),HIP出现时间推迟(P<0.01),HIP出现率降低(P<0.05)。结论在缺氧过程中应用10μmol·L-1的丙泊酚对缺氧无糖损伤海马脑片具有保护作用。Objective To investigate the protective effect of propofol on injury induced by oxygen glucose deprivation(OGD)in rat hippocampal slices.Methods Ischemia in vitro was induced by exposing slice to artificial cerebrospinal fluid(aCSF),from which oxygen andglucose were deprivated and gassed with 95%N_2/5%CO_2.Propofol(1,10 and 20 μmol·L~ -1)and intralipid were administrated before OGD(pretreatment group)or during OGD(treatment group),respectively.Orthodromic population spikes(OPS)and hypoxic injury potential(HIP)in CA1 area of rats in hippocampal slices were recorded by extracellular microelectrode recording technique.Results In treatment group,10 μmol·L~ -1 propofol significantly delayed the disappearance time of OPS after OGD(P<0.05),improved the recovery rate of OPS after reoxygenation(P<0.05),delayed the appearance time of HlP after OGD(P<0.05)and reduced the appearance rate of HIP(P<0.05)when compared with control.Conclusion Propofol(10 μmol·L~ -1)administrated during OGD has protective effect on injury induced by OGD in rat hippocampal slices.
