摘 要:目的 研究PTEN对血管紧张素Ⅱ(AngⅡ)所致心肌细胞肥大的影响。方法 构建携带PTEN基因的腺病毒载体,感染原代培养的乳鼠心肌细胞,用AngⅡ作为肥大刺激因素,测定ANF、βMHC与心肌细胞大小。结果 对照组与仅携带GFP的腺病毒感染的心肌细胞在AngⅡ作用下,ANF、βMHC表达与细胞直径显著增高,而PTEN过度表达则对上述改变具有抑制作用。结论 PTEN能够负性调控AngⅡ所致的心肌细胞肥大。Objective To study the effects of PTEN on angiotensin-Ⅱ (Ang-Ⅱ) induced cadiomyocytic hypertrophy. Methods Wild type PTEN gene was transferred into the primary generation of cardiomyocytes from neonatal rats through the recombinant adenovirus containing PTEN gene. Then the cardiomyocytes were irritated with Ang-Ⅱ. ANF, MHC and the size of the cardiomyocytes, which served as the index of hypertrophic response, were determined. Results After Ang-Ⅱ stimulation, the level of ANF and MHC and the cardiomyocytic size were significantly increased in the cardiomyecytes with normal expression of PTEN but the hypertrophic response was markedly suppressed in those with high expression of PTEN. Conclusion PTEN can down-regulate the hypertrophic response of cardiomyocytes induced with Ang-Ⅱ.
