摘  要:本文简要阐述了RAPD、RFLP、AFLP、SSR等几种主要的建立DNA指纹图谱的分子标记技术及其在杂交水稻种子纯度和真伪鉴定中的研究和应用情况,同时评述了这几种分子标记技术各自的优缺点,并就水稻DNA提取方法的改进、PCR扩增反应程序和反应体系的优化、扩增产物的检测以及DNA指纹在杂交稻种子纯度和真伪鉴定中的准确性等问题进行了讨论。DNA指纹在杂交水稻种子纯度和真伪鉴定中具有广阔的应用前景,关键是要建立一套完善的、标准化的技术体系,进一步简化实验操作程序,降低成本,真正做到简单快速、准确可靠,使DNA指纹能用于规模化、商业化的种子质量鉴定之中。In this paper, several types of DNA molecular markers of being employed to construct DNA fingerprinting are introduced, such as RAPD, RFLP, AFLP and SS R. The study and application of these molecular markers in hybrid rice seed puri ty and authenticity identification are surveyed and their advantages and disadva ntages are analysed. This paper discusses the DNA extraction methods, the progra m and volumes of PCR reaction, the detection of PCR products and the acuteness o f DNA fingerprinting identification. The key that application of DNA fingerprint ing in hybrid rice seed purity and authenticity identification would have wide p rospect is to construct an empirical and standard technique. And the method shou ld be further simplified and the costs should be reduced in order to reach the g oal of automation and commercialization of seed identification.
