中药金叶败毒制剂体外抗人巨细胞病毒作用观察

摘　　要：目的:研究中药金叶败毒制剂(JYBD)抗人巨细胞病毒(HCMV)感染的可能机制。方法:采用体外细胞培养技术建立HCMV感染细胞模型,应用更昔洛韦(GCV)和JYBD进行干预,采用定量RT-PCR法检测干预后不同时间感染细胞HCMV即刻早期基因IE86mRNA的表达水平,观察感染细胞的病变进展情况,同时采用放射免疫法测定受染细胞细胞因子TNF-α和IL-6的分泌水平。结果:JYBD与GCV对HCMVIE86mRNA的表达以及HCMV所致细胞病变有明显的抑制作用,降低受染细胞TNF-α、IL-6的分泌水平,JYBD作用弱于GCV。结论:JYBD可通过抑制HCMVIE基因的转录,促进细胞因子的分泌而发挥体外抗HCMV作用。Aim: To study the antiviral mechanism of the Chinese traditional herb Jinyebaidu (JYBD)on human cytomegalovirus(HCMV) infection. Methods: HCMV infected cell model was established by coincubating the human embryo lung cells and HCMV AD169 in vitro, and the expression of the HCMV immediate early gene 86 mRNA(HCMV IE86 mRNA) in infected cells was measured by using quantitative RT-PCR and the development of cytopathic effect(CPU) caused by HCMV was observed before and after being treated with ganciclovir(GCV) and JYBD, respectively. The levels of tumor necrotic factor alpha(TNF-α) and interleukin-6(IL-6)in cultured cell supernatant were detected using radioimmunoassay at different time points. Results: JYBD and GCV had strong inhibitive effect on the expression of HCMV IE86 mRNA and the CPU.JYBD and GCV could decrease the secretion of TNF-α and IL-6 in the infectious cells. Conclusion: JYBD can enhance the immunity and has obvious inhibitive effect on HCMV in vitro.
